What is SLU-PP-332?
SLU-PP-332 is a synthetic small-molecule agonist of the estrogen-related receptors (ERRs), a family of nuclear receptors closely related to estrogen receptors but which do not bind estrogen itself. It is a pan-ERR agonist, meaning it activates all three isoforms: ERRα (strongest potency: EC₅₀ ≈ 98 nM) ERRβ (EC₅₀ ≈ 230 nM) ERRγ (EC₅₀ ≈ 430 nM) Developed by researchers at Washington University in St. Louis (SLU) and collaborators, it is often described as an “exercise mimetic” because it activates metabolic pathways normally triggered by aerobic/endurance exercise, particularly through upregulation of PGC-1α (a master regulator of mitochondrial biogenesis and energy metabolism). It is not a peptide (despite some vendors mislabeling it as such) — it is an oral small molecule (CAS 303760-60-3). How Does It Work? ERRs (especially ERRα) control genes involved in mitochondrial function, fatty acid oxidation, glucose metabolism, and muscle fiber adaptation. During exercise, ERR activity increases naturally. SLU-PP-332 binds and activates ERRs, mimicking this response: Boosts mitochondrial biogenesis and respiration in skeletal muscle. Shifts muscle fibers toward fast oxidative (Type IIa) — more fatigue-resistant and efficient at burning fat. Increases energy expenditure and fat oxidation without increasing physical activity or suppressing appetite. Unlike GLP-1 drugs (e.g., semaglutide/Ozempic), it does not reduce food intake — weight loss comes purely from metabolic reprogramming.
